Indicator displacement assays that detect bilayer membranes enriched in phosphatidylserine

چکیده
Three indicator displacement assays are described for the detection of phosphatidylserine in a bilayer membrane. A series of Zn-dipicolylamine coordination compounds are used to bind selectively to the phosphatidylserine and act as a colorimetric chemosensing ensemble when combined with the UV-Vis indictor pyrocatechol violet. A similar displacement assay uses a coumarin methylsulfonate derivative as a fluorescent indicator, and a third assay involves quenching of calcein fluorescence by Cu and subsequent fluorescence restoration upon addition of phosphatidylserine. In the best case, vesicle membranes containing as little as 5% phosphatidylserine could be detected under physiologically relevant conditions using as little as 10 mM sensing ensemble, and two of the three systems allow vesicles containing 50% phosphatidylserine to be detected by the naked eye.

Active mechanisms are needed to describe cell responses to submicrosecond, megavolt-per-meter pulses: cell models for ultrashort pulses.
Intracellular effects of submicrosecond, megavolt-per-meter pulses imply changes in a cell's plasma membrane (PM) and organelle membranes. The maximum reported PM transmembrane voltage is only 1.6 V and phosphatidylserine is translocated to the outer membrane leaflet of the PM. Passive membrane models involve only displacement currents and predict excessive PM voltages (approximately 25 V). Her...

A Zn-dipicolylamine coordination compound is shown to sense the presence of anionic phospholipids in a membrane bilayer. The sensor contains two dipicolylamine subunits attached to an anthracene scaffold, which exhibits a maximum absorbance at 380 nm, and undergoes an enhancement in fluorescence intensity when exposed to membranes enriched in phosphatidylserine. For these reasons, the compound ...
متن کاملNaja naja oxiana Cobra Venom Cytotoxins CTI and CTII Disrupt Mitochondrial Membrane Integrity: Implications for Basic Three-Fingered Cytotoxins
Cobra venom cytotoxins are basic three-fingered, amphipathic, non-enzymatic proteins that constitute a major fraction of cobra venom. While cytotoxins cause mitochondrial dysfunction in different cell types, the mechanisms by which cytotoxins bind to mitochondria remain unknown. We analyzed the abilities of CTI and CTII, S-type and P-type cytotoxins from Naja naja oxiana respectively, to associ...
متن کاملFacilitated phosphatidylserine flip-flop across vesicle and cell membranes using urea-derived synthetic translocases.
Tris(2-aminoethyl)amine derivatives with appended urea and sulfonamide groups are shown to facilitate the translocation of fluorescent phospholipid probes and endogenous phosphatidylserine across vesicle and erythrocyte cell membranes. The synthetic translocases appear to operate by binding to the phospholipid head groups and forming lipophilic supramolecular complexes which diffuse through the...
متن کاملPurification, and Functional Reconstitution of the P4- Atpase, Atp8a2, a Phosphatidylserine Flippase in Photoreceptor Disc Membranes
P4-ATPases comprise a relatively new subfamily of P-type ATPases implicated in the energy dependent translocation of aminophospholipids across cell membranes. In this study we report on the localization and functional properties of Atp8a2, a member of the P4-ATPpase subfamily that has not been studied previously. RT-PCR revealed high expression of Atp8a2 mRNA in the retina and testis. Within th...
